Adsorption and release studies of new cephalosporin from chitosan-g-poly(glycidyl methacrylate) microparticles

Porous microparticles of chitosan-g-poly(glycidyl methacrylate) were obtained by grafting chitosan onto crosslinked networks based on glycidyl methacrylate and ethylene glycol dimethacrylate using suspension polymerization technique. A new cephalosporin from the indazole class prepared by acylation of 7-Aminodesacetoxycephalosporanic acid with mixed anhydride of 5-nitroindazole-1-yl-acetic acid was used as active principle. The cephalosporin-microparticle systems were characterized by FT-IR spectroscopy, SEM and AFM analysis. Batch experiments were carried out to study the influence of initial drug concentration, temperature, contact time, drug:microparticles ratio and pH on the adsorption process of cephalosporin onto porous crosslinked microparticles. Two-parameter and three-parameter isotherm models were used to evaluate the adsorption equilibrium. The values of diffusion coefficients indicate that the cephalosporin adsorption onto microparticles was controlled by both film diffusion and pore diffusion mechanisms. The analysis of the kinetic data of the release process indicate that the release mechanism of cephalosporin from microparticles corresponds to the anomalous transport mechanism.

Pore size tailoring from ultrafiltration to nanofiltration with PVC-g-PDMA via rapid immersion thermal annealing

Different from the typical interfacial polymerization for nanofiltration membrane fabrication, we proposed a novel method to prepare charged nanofiltration membrane via rapid immersion thermal annealing process with the ultrafiltration membrane, which was prepared from the amphiphilic copolymer, poly(vinyl chloride)-g-poly(2-(dimethylamino)ethyl methacrylate) (PVC-g-PDMA) via non-solvent induced phase separation. The pore structures and chemical properties of the membranes, and annealing conditions including the media where the membrane was annealed, annealing temperature and time, were investigated in detail. It is found that the porous separation layer was collapsed to be a stable, defect-free, and smooth film as the dense separation layer of nanofiltration membrane when it was annealed in n-heptane above its glass transition temperature (Tg) for only 1 min while the pore size cutoff was tailored from 20 nm to 2 nm. Salt rejections were in the order of MgCl2 > NaCl > MgSO4 > Na2SO4 at neutral condition due to Donnan repulsion, indicating a typical positively charged nanofiltration membrane. Overall, this rapid immersion thermal annealing process provides a facile and effective strategy to tailor ultrafiltration membrane toward nanofiltration applications for amphiphilic copolymer based membranes.

Accurate chiral pattern recognition for amines from just a single chemosensor

The current work proposes a novel determination method for enantiomeric excess (ee) in (mono- and di-) amines using molecular self-assembly. A pyridine-attached binaphthyl derivative ((R)-1) exhibits fluorescence responses based on imine formation between the aldehyde group of (R)-1 and target chiral amines (i.e. cyclohexane diamine (CHDA), 2-amino-1,2-diphenylethanol (ADPE), 1,2-diphenylethylenediamine (DPDA), 1-amino-2-indanol (AID), and leucinol) in the presence of zinc(ii) ions (Zn2+). Because of the multi-optical responses which are derived from the variation of chiral complexes, pattern recognition-based discrimination (i.e. linear discriminant analysis (LDA)) has been achieved for five types of enantiomeric pairs of amines. Possessing such a discrimination capability in combination with data processing (LDA and an artificial neural network) allows accurate determination (prediction error < 1.8%) of the % ee of individual targets such as CHDA which is one of the main components of pharmaceutical drugs. The simple molecular self-assembled system enabled simultaneous multi-chiral discrimination and % ee determination of unknown samples.
